Webinar Abstract: 

Endosomal sequestration of lipid-based nanoparticles (LNPs) remains a formidable barrier to intracellular delivery of mRNA. Structure-activity analysis of cholesterol analogues reveals that incorporation of C-24 alkyl phytosterols into LNPs (eLNPs) enhances gene transfection. eLNPs show a polyhedral structure and 3D single-particle tracking shows enhanced intracellular diffusivity of eLNPs relative to LNPs, and high Galectin 8 recruitment onto the endosome suggesting enhanced escape. I will briefly discuss extrahepatic gene delivery of LNPs for the treatment of cystic fibrosis, retinal degeneration and COVID-19 therapeutics. Deconvolution of LNP surface composition and structural properties of nanoparticles, and their subcellular interactions can enable designs to boost intracellular delivery of mRNA, especially in non- hepatic targets.
